摘要:华重楼(Paris polyphylla Smith var.chinensis (Franch.)Hara)二级濒危物种,2020年版《中国药典》收载的重楼药材2 种基原植物之一。为了解决华重楼优良种苗繁育问题,以华重楼顶芽为材料,研究外植体灭菌、愈伤组织诱导、愈伤组织分化、瓶内生根以及移栽驯化,开展华重楼高效组培快繁体系建立及优化研究。结果表明:(1)消毒最佳方法是以体积浓度为75% 的乙醇浸泡60 s后再以质量浓度为0.1% 的HgCl2 消毒10 min,成功率为35.27%。(2)无菌苗叶诱导愈伤组织的最佳配方为MS+ 2.0 mg?L–1 6-BA + 0.5 mg?L–1 2,4-D+ 0.5 mg?L–1 IBA,培养30 d ,诱导率93.33%,绿色松散,分化能力强;无菌苗茎段诱导愈伤组织的最佳配方为MS+ 2.0 mg?L–16-BA + 1.5 mg?L–1 2,4-D + 0.5 mg?L–1 IBA,培养30 d ,诱导率91.67%,绿色紧密,分化能力强。(3)诱导愈伤组织分化最佳培养基为MS+ 1.5 mg?L–1 6-BA + 2.0 mg?L–1 ZT + 0.6 mg?L–1 IBA,芽分化率96.67%,增值系数为19/30 d。(4)生根最佳培养基为MS+ 1.5 mg?L–1 6-BA + 0.5 mg?L–1 IBA,生根率为100%,平均生根20.12条。(5)生根苗在小颗粒泥炭土中,成活率达到92.30%。该研究建立了华重楼高效组培快繁体系,为华重楼优质种苗工厂化生产奠定良好基础。 |

Abstract: Paris polyphylla Smith var.chinensis (Franch.)Hara is a class II endangered species. It is one of the two original plants of Paris polyphylla recorded in Chinese Pharmacopoeia (2020 Edition). In order to solve the problem of breeding excellent seedlings of Paris polyphylla Smith var.chinensis (Franch.)Hara,the research is conducted on the establishment and optimization of high-efficient tissue culture and rapid propagation system of Paris polyphylla Smith var.chinensis (Franch.)Hara by taking apical bud induced aseptic seedling as the material. Explant sterilization, callus induction, callus differentiation, test-tube rooting and transplanting and domestication were studied. The results are: (1) The best ratio of sterilization was 60 seconds of 75% alcohol + 10 minutes of 0.1% HgCl2, and the pollution rate was 35.27%. (2) The best formula for callus induction by aseptic seedling leaf is MS + 2.0 mg?L-1 6-BA + 0.5 mg?L-1 2,4-D + 0.5 mg?L-1 IBA, for 30 days, and the induction rate was 93.33% with diffuse green and strong differentiation potential. The best formula for callus induction by aseptic seedling shoot MS+ 2.0 mg?L-1 6-BA + 1.5mg.L-1 2,4-D + 0.5 mg.L-1 IBA, for 30 days, and the induction rate was 91.67%. (3) The most suitable medium for induction of the callus differentiation was MS + 1.5 mg?L-1 6-BA + 2.0 mg?L-1 ZT + 0.6 mg?L-1 IBA, with 19 as the multiplication coefficient. (4) The most suitable medium for rootage was MS + 1.5 mg?L-1 6-BA + 0.5 mg?L-1 IBA, achieving 100% rootage rate with 20.12 of the average number. (5) Plantlets were transplanted to small particle peat soil, and 92.30% rooted plants survived. The research has established the high-efficient tissue culture and rapid propagation system of Paris polyphylla Smith var.chinensis (Franch.)Hara, which can be applied in mass production and play a foundation for providingseedlings and factory production |
