为研究白花银背藤(Argyreia seguinii Levl.)生物碱类成分及其细胞毒活性,该文采用硅胶、ODS、Sephadex LH-20及半制备HPLC等柱色谱方法对白花银背藤的75%乙醇提取物进行分离纯化,根据理化性质及波谱数据鉴定化合物结构;通过MTS法检测化合物对5种肿瘤细胞的增殖抑制活性。结果从白花银背藤中分离得到9个生物碱类化合物,分别鉴定为N-反式桂皮酰对羟基苯乙胺(1)、N-反式对香豆酰基酪胺(2)、N-反式阿魏酰酪胺(3)、N-顺式阿魏酰酪胺(4)、1H-吲哚-3-甲醇(5)、金色酰胺醇酯(6)、8-oxotetrahydropalmatine(7)、8-oxypalmatine(8)和12,13-dihydro-8-oxoberberine(9),其中化合物1、4–9均为首次从该植物中分离得到;化合物7和8对肝癌细胞SMMC-7721显示细胞毒活性,IC50分别为(15.50 ± 0.76)μmol·L-1和(14.24 ± 0.72)μmol·L-1。 |

To study the alkaloids from A. seguinii and their cytotoxic activity, the 75% ethanol extract from A. seguinii was isolated and purified by silica gel, ODS, Sephadex LH-20 and semi-preparative HPLC, and the structures of obtained compounds were identified by physicochemical properties and spectral data. The cytotoxic activity of the compounds were determined by MTS method. Nine compounds were isolated and identified as trans-N-cinnamoyltyramine (1), trans-N-p-coumaroyltyramine (2), N-trans-feruloyltyramine (3), N-cis-feruloyltyramine (4), 1H-indole-3-carbaldehyde (5), aurantiamide acetate (6), 8-oxotetrahydropalmatine (7), 8-oxypalmatine (8) and 12,13-dihydro-8-oxoberberine (9). Compounds 1, 4–9 are isolated from this plant for the first time, and compounds 7 and 8 showed inhibitory activity against the cancer cells SMMC-7721 with the IC50 values of (15.50 ± 0.76) μmol·L-1 and (14.24 ± 0.72) μmol·L-1, respectively. |
