| 厚果崖豆藤化学成分及其对MDA-MB-231细胞的抑制活性 |

| 为探究厚果崖豆藤化学成分及其对MDA-MB-231细胞的抑制活性,该研究采用硅胶、Sephadex LH-20对厚果崖豆藤提取物进行分离纯化,根据理化特性和波谱数据鉴定化合物的结构,并采用MTT法测试化合物对MDA-MB-231细胞增殖的抑制活性。结果表明:(1)从厚果崖豆藤中分离鉴定出22个化合物,分别为mtricrin(1)、4-羟基-3-甲氧基苯乙酮(2)、6-甲氧基-7-羟基香豆素(3)、globuxanthone(4)、对甲氧基苯甲酸(5)、oxophoeine(6)、isoshonnin(7)、schizndriside(8)、nudiposide(9)、rtemetin(10)、 hentricontnol(11)、clliphyllin(12)、corymoside(13)、norrchycourmrin(14)、2, 5-二羟基苯乙酮(15)、2, 4-二羟基苯乙酮(16)、mkoline(17)、5, 6, 7-三甲氧基香豆素(18)、5, 7, 4'-三羟基-3', 5'-二甲氧基黄酮(19)、5, 4'-二羟基-3, 7, 3'-三甲氧基黄酮(20)、5, 4'-二羟基-7-甲氧基黄酮(21)、3, 5, 7-三羟基-6, 8-二甲基黄酮(22)。所有化合物均为首次从厚崖豆藤中分离鉴定出。(2)化合物6和化合物11对MDA-MB-231细胞增殖具有良好的抑制活性,其中化合物6的IC50值为5.71 μmol·L-1,与阳性药物5-氟尿嘧啶相当(IC50值为5.53 μmol·L-1)。该研究结果揭示了厚果崖豆藤的化学成分,为深入开展厚果崖豆藤抑制MDA-MB-231细胞增殖研究提供了科学依据。 |

| In order to investigate the chemical constituents of Millettia pachycarpa and their inhibitory activity on MDA-MB-231 cells, the extract of M. pachycarpa was isolated and purified by silica gel and Sephadex LH-20. The structures of the compounds were identified according to the physicochemical properties and spectral data. The inhibitory activities of the compounds on MDA-MB-231 cells were tested by MTT assay. The results were as follows:(1)Twenty-two compounds were identified from M. pachycarpa as mtricrin(1), 4-hydroxy-3-methoxycetophenone(2), 6-methoxy-7-hydroxycoumrin(3), globuxanthone(4), p-methoxybenzoic acid(5), oxophoeine(6), isoshonnin(7), schizndriside(8), nudiposide(9), rtemetin(10), hentricontnol(11), clliphyllin(12), corymoside(13), norrchycourmrin(14), 2, 5-dihydroxyacetophenone(15), 2, 4-dihydroxyacetophenone(16), mkoline(17), 5, 6, 7-trimethoxycoumarin(18), 5, 7, 4' -trihydroxy-3', 5' -dimethoxyflavonone(19), 4', 5-dihydroxy-3, 3', 7-trimethoxyflavonone(20), 5, 4'-dihydroxy-7-methoxyflavonone(21), 3, 5, 7-trihydroxy-6, 8-dimethyl flavone(22). All the compounds were identified from M. pachycarpa for the first time.(2)Compounds 6 and 11 showed good inhibitory activities on the proliferation of MDA-MB-231 cells. The IC50 value of Compound 6 was 5.71 μmol·L-1, which was similar to that of 5-fluorouracil(IC50 value was 5.53 μmol·L-1). The results of this study reveal the chemical constituents of M. pachycarpa, and provide a scientific foundation for further research on inhibiting the proliferation of MDA-MB-231 cells. |
